Keeping a Physarum polycephalum culture usually comes down to a few basics: a damp surface, modest feeding, clean handling and close observation. Active cultures can change shape quickly, then slow down or form a resting structure when conditions shift, so the best care routine is one that keeps the surface usable without flooding it.

Moisture, food and surface care

Rolled oats are commonly used because they are easy to place, watch and remove. Add a small amount near the active edge, then wait for colonisation before feeding again. Avoid covering the whole surface with food, and remove heavily degraded or persistently uneaten oats when practical. Overfeeding can leave residue behind and encourage unwanted microbial growth.
Plain water agar can be useful as a hydrated surface, but it is usually a substrate rather than the main food source. If the plate dries, cracks or becomes difficult to maintain, move a small piece of active material onto a fresh moist surface with food nearby.
Reading the culture
Active Physarum polycephalum may appear as a yellow sheet, fan, vein network or branching front. Colour and shape vary with light, substrate, food and culture condition, so appearance alone should not be treated as a full diagnosis. Change over time is often more useful than a single photograph.
If the culture seems thin, slow or inactive, compare it over time before assuming it has failed. Dryness, depleted food, unsuitable temperature or a resting state can all change how it looks. Fuzzy or filamentous growth is more consistent with fungal contamination than with active plasmodium, and old food can soften, darken and support microbial growth.
Resting states and transfers
When conditions become unsuitable, the culture may form a sclerotium. That resting form can look dry, crusted or vein-like, and it is not the same as an actively feeding plasmodium. If viable material is present, rehydration may help, but recovery is not guaranteed and revival time varies.
For transfers, move a small piece of active plasmodium or colonised substrate onto a fresh moist surface with food nearby. Use clean tools, minimise handling and keep a backup where practical. If unwanted growth is localised, transfer visibly healthy material away from the affected area rather than trying to rescue the whole plate at once.
